氯仿-甲醇法和酸水解法测定禽蛋中脂肪的方法比较
目的 对比氯仿-甲醇法和酸水解法测定富含磷脂等结合态脂肪的禽蛋制品中脂肪含量的方法。方法 采用氯仿-甲醇提取法对比GB 5009.6-2016《食品安全国家标准 食品中脂肪的测定》中规定的禽蛋制品脂肪测定方法-酸水解法, 测定禽蛋制品中的脂肪含量, 考察了2种方法测定不同蛋制品的提取结果、提取时间、精密度和重复性。结果 氯仿-甲醇提取法可以有效地提取蛋制品的磷脂等结合态脂肪, 测定结果更接近于理论值, 而酸水解法测定结果普遍偏低。氯仿-甲醇提取法提取需要至少1.8 h, 酸水解实验提取过程需要至少 2.5 h, 氯仿-甲醇提取法能缩短实验周期。氯仿-甲醇提取法对禽蛋制品脂肪的测定具有较高的精密度和较好的重复性, 相对标准偏差在0.91%~1.62%之间, 而酸水解的相对标准偏差在6.41%~9.13%之间。结论 氯仿-甲醇法弥补了酸水解法水解磷脂的不足且测定快速、准确, 适合测定禽蛋制品中的脂肪含量。
Objective To compare the methods of chloroform methanol method and acid hydrolysis method for the determination of fat content in egg products rich in phospholipid and other conjugated fats. Methods The method of chloroform-methanol extraction was used to determine the fat content in egg products, and compared with the method of acid hydrolysis stipulated in GB 5009.6?2016 National food safety standard-Determination of fat in foods. The results, extraction time, precision and repeatability of the 2 methods were investigated. Results Chloroform-methanol extraction method could effectively extract phospholipid and other bound fats in egg products, and the determination results were closer to the theoretical values, while the determination results by acid hydrolysis method were generally low. Chloroform-methanol extraction took 1.8 h at least, acid hydrolysis experiment took 2.5 h at least, and chloroform-methanol extraction could shorten the experimental period. Chloroform-methanol extraction method had high precision and good repeatability in the determination of egg fat, and the relative standard deviation was 0.91%?1.62%, while the relative standard deviation of acid hydrolysis was between 6.41%?9.13%. Conclusion Chloroform-methanol method is fast and accurate, and is suitable for the determination of fat content in egg products, which makes up the deficiency of acid hydrolysis method.
