不同产地马齿苋高效液相指纹图谱的建立及其聚类分析
目的 建立不同产地马齿苋的高效液相(high performance liquid chromatography, HPLC)指纹图谱并进行聚类分析。方法 采用资生堂CAPCELL PAK ADME(250 mm×4.6 mm, 5 μm)为色谱柱; 检测波长为 320 nm; 柱温设置为30 ℃; 流速为1 mL/min; 流动相为乙腈以及0.1%磷酸水溶液, 梯度洗脱; 通过相似度评价、聚类分析对指纹图谱进行评价。结果 建立的指纹图谱共标定了26个共有峰, 各产地不同批次样品指纹图谱相似度均在0.9以上, 以共有峰峰面积的聚类分析可以较好的区分不同产地样品。结论 该方法操作简便、准确可靠、重复性好, 普适性高, 为马齿苋质量标准的提高提供了参考。
Objective To establish high performance liquid chromatography (HPLC) fingerprints of Portulaca oleracea from different regions and perform cluster analysis. Methods Shiseido CAPCELL PAK ADME (250 mm×4.6 mm, 5 μm) was used as the chromatographic column; the detection wavelength was 320 nm; the column temperature was 30 ℃; the flow rate was 1 mL/min; the mobile phase was acetonitrile and 0.1% phosphoric acid aqueous solution (gradient elution); The fingerprints were evaluated by cluster analysis and similarity evaluation. Results A total of 26 common peaks were calibrated by the established fingerprint, and the similarity of the fingerprints of different batches of samples from different regions were above 0.9. Cluster analysis based on the area of common peaks can better distinguish samples from different regions. Conclusion The method is simple, accurate, reliable, reproducible, and universal. It provides a reference for improving the quality standard of Portulaca oleracea.
