Real-time环介导等温扩增检测枣花蜜源成分
目的 建立real-time环介导等温扩增技术(loop-mediated isothermal amplification, LAMP)检测枣花蜜源成分的方法。方法 以枣转录间隔区(internal transcribed spacer, ITS)为靶序列, 用软件Primer Explorer V5与软件Oligo 7设计LAMP特异性引物, 优化反应温度, 测试灵敏度、特异性和适用性。结果 所建立的方法在反应温度55 ℃条件下, 引物特异性强, 最低检测限为≥1 pg/μL大枣DNA。结论 该方法具有特异性强、灵敏度高、操作简单等优势, 适合商业枣花蜜掺假快速检测。
Objective To establish a real-time loop-mediated isothermal amplification (LAMP) method for the detection of Fructus jujubae nectar. Methods The LAMP-specific primers were designed with Primer Explorer V5 and Oligo 7 to optimize the reaction temperature, sensitivity, specificity and applicability using the internal transcribed spacer (ITS) as the target sequence. Results The established method was of high specificity under the condition of reaction temperature of 55 ℃, and the detection limit was≥1 pg/μL genomic DNA of jujube. Conclusion The method has the advantages of high specificity, high sensitivity and simple operation, which is suitable for rapid detection of jujube nectar adulteration in commercial.
