探索7.5%氯化钠肉汤增菌液OD值与生物量变化关系
目的 通过OD值的测定,确定一种简单、快捷、准确的方法判定液体培养基的增菌效果。方法 本实验分别通过在200、350、470、550、660 nm5种不同波长下测定培养基空白及增菌液OD值,选择550 nm作为实验测定波长。通过测定不同初始菌数、不同培养时间菌悬液550nm 的OD值以及菌落总数,建立回归性方程。结果 4株标准菌株均表现出当OD值低于0.11时与菌落总数的对数值呈现正相关,且r2>0.99。当OD值大于0.11时,菌悬液中含有菌落数均达到107以上,菌株不再以对数形式进行繁殖代谢,4株菌均呈现出线性相关性,且r2>0.95。结论 对于7.5%氯化钠肉汤培养基可选用550 nm的吸光值对培养基增菌效果进行判定,以得到一种更加准确、快速、高效的评价液体培养基质量的方法。
Objective The purpose of this study was to to determine a simple, fast and accurate method to determine the effect of adding bacteria in liquid culture medium by the determination of OD value. Method In this study, the medium blank and the OD value of bacterial culture solution were measured in five different wavelengths:The experiment measured the medium blank and the OD value of the cultivation at five different wavelengths: 200 nm, 350 nm, 470 nm, 550 nm, and 660 nm and 550nm was selected as the experimental wavelength. The regression equation was established by measuring the OD value of bacteria at 550 nm and the total number of colonies at different initial bacterial counts, different culture times.Results: All the four standard strains showed a positive correlation with the total number of colonies when the OD value was lower than 0.11, and r2> was 0.99. When the OD value was greater than 0.11, the bacterial suspensions contained more than 107 colonies, and the bacterial strains were no longer capable of reproduction and metabolism in logarithmic form. The numerical value and OD value results were analyzed by the total number of colonies, and all the four strains presented linear correlation, with r2>0.95. Conclusion: For 7.5% sodium chloride broth medium, 550nm light absorption value could be used to determine the effect of bacteria growth.Key words: 7.5 % sodium chloride broth; Staphylococcus aureus ;OD value; total number of colonies
