外源乙烯对马铃薯萌芽过程中酚类物质代谢的影响
目的 探究外源乙烯对马铃薯萌芽过程中酚类物质代谢的影响。方法 设立4 个处理: M0 为清水清洗处理, M1、M2、M3 分别为在水中将1、2、3 包乙烯固体释放剂浸泡, 对休眠期已过的马铃薯块茎进行处理。采用Folin 酚法、pH 示差法等方法, 定期测定总酚、黄酮、花青素含量的变化, 并通过高效液相色谱法对酚类物质进行提取和分析。结果 与对照组相比, 外源乙烯对马铃薯酚类物质有明显的影响, 可明显抑制马铃薯块茎中总酚的积累, 且1 包固体乙烯释放剂(2.5 g)的效果更明显; 马铃薯块茎中总黄酮积累速度明显减慢, 3包固体乙烯释放剂(7.5 g)的效果则更明显。另外, 花青素含量会因外源乙烯的处理而降低, 尤其是3 包固体乙烯释放剂(7.5 g)处理组。外源乙烯对酚类组分的影响主要表现在咖啡酸、香草酸以及槲皮素上, 而对其他组分影响较小。结论 外源乙烯抑制马铃薯发芽的过程中, 酚类物质代谢受到了影响, 总酚、总黄酮的积累速度发生明显的降低。
Objective To explore the effect of exogenous ethylene on the metabolism of phenolic substances in potato germination. Methods Four treatments were set up: M0 was water treatment group and M1, M2, M3 were groups which soaked 1, 2, and 3 vinyl solid release agents in water, respectively, treating the potato tubers over thedormant period. The changes of the content of total phenol, flavone and anthocyanin were determined by Folin phenol method and pH differential method. The changes of anthocyanin content in total phenol flavonoids were measured regularly, and the phenol substances were extracted and analyzed by high performance liquid chromatography. Results Compared with the control group, exogenous ethylene had obvious influence on the potato phenolic substances, which could obviously inhibit the accumulation of total phenol in potato tubers. And the effect of 1 pack of solid ethylene releasing agent (2.5 g) was more obvious. The accumulation rate of total flavonoids in potato tubers was significantly suppressed and the effect of 3 packs of solid ethylene releasing agent (7.5 g) was more obvious. However, anthocyanin content could be reduced by the treatment of exogenous ethylene, especially 3 packs of solid ethylene release agent (7.5 g) treatment group. The effects of exogenous ethylene on phenolic components were mainly reflected in caffeic acid, vanillic acid and quercetin, and the effects on other components were less. Conclusion The metabolism of phenolic substances is affected by exogenous ethylene in the process of inhibiting the germination of potato, the accumulation rate of total phenol and total flavonoids is significantly reduced.
