养生醋中香茅醇和香叶醇含量分析
目的 建立养生醋中香茅醇和香叶醇含量的测定方法。方法 正己烷提取后, 用安捷伦HP-FFAP 19091-105色谱柱分离, 气相色谱氢火焰离子化检测器(flame ionization detector, FID)检测。在优化的气相色谱分析条件下, 选择高纯氮气为载气, 流速为1.5 mL/min, 进样口温度为260 ℃, 设程序初始温度为60 ℃, 持续1 min, 以2 ℃/min升温至230 ℃并持续10 min。结果 香茅醇、香叶醇与基质分离良好, 其含量在0.005~1 g/L浓度范围内线性关系良好, 相关系数分别为0.9995和0.9994; 在0.2 mg/L的添加水平时, 香茅醇和香叶醇的平均回收率为92.3%和97.5%; 1 mg/L添加水平时, 香茅醇和香叶醇的平均回收率为95.1%和95.6%; 50 mg/L添加水平时, 香茅醇和香叶醇的平均回收率为92.8%和94.0%, RSD为0.32%~1.06 %和0.46%~2.33%(n=5)。采用该方法对实际养生醋产品进行含量分析, 香茅醇含量为9.90 mg/L, 香叶醇含量为5.85 mg/L。结论 建立的方法灵敏、准确、可靠, 分析时间短, 抗干扰性好, 可用于企业产品开发研究及企业产品质量控制。
Objective To establish the method for the content determination of citroniol and geraniol in healthy vinegar. Methods The sample was extracted from n-hexane. Chromatographic column was Agilent technologies HP-FFAP 19091-105 and the sample was detected by gas chromatography with flame ionization detector. Under the conditions of optimization of gas chromatography, high purity nitrogen was chosen as the carrier gas, flow rate was 1.5 mL/min, the sample temperature was 260 ℃, the initial temperature was 60 ℃, and last 1 min, with 2 ℃/min and temperature to 230 ℃ for 10 min. Results Citronellin and geraniol were separated well from the matrix, and they had good relationships within the concentration range of 0.005-1 g/L(r=0.9995 and 0.9994). The average recoveries of ctroniol and graniol were 92.3% and 97.5% at spiked level of 0.2 mg/L, 95.1% and 95.6% at spiked level of 1 mg/L and 92.8% and 94.0% at spiked level of 50 mg/L, with relative standard deviations (RSD) of 0.32%-1.06% and 0.46%-2.33% (n=5). The content analysis of healthy vinegar was done with this method and the contents of citronol and geraniol were 9.90 mg/L and 5.85 mg/L. Conclusion This method is sensitive, accurate and reliable with short analysis time and good anti-interference, which can be used for enterprise product development research and enterprise product quality control.
