Objective To discuss the reasonability of the limit of detection in standard of SN/T3730.4-2013 Identification of domestic animal ingredient in food and feed-Part 4: Detection of donkey-derived ingredient-real-time PCR method. Methods According to the primers and probe sequences provided by SN/T 37344-2013, the authenticity of sampling donkey meat in the market was identified by real-time fluorescence PCR detection method. Results The samples with Ct value between 25 and 35 should be judged as the donkey component detected in accordance with SN/T 37344-2013 standard. By amplifying and sequencing the above samples with equine primers, it was found that the samples were horse meat instead of donkey meat. Conclusion It is suggested that different limits of detection should be set according to different inspection objects, so as to reduce the probability of false positive.
