优化EPA 1623法检测生活饮用水中隐孢子虫和 贾第鞭毛虫
目的 建立提高生活饮用水中隐孢子虫和贾第鞭毛虫(两虫)检出率的方法。方法 用1.2 μm微孔滤膜通过真空抽滤的方式过滤两虫加标样品, 后将滤膜置于磷酸缓冲盐溶液(phosphate buffer saline, PBS)中进行超声洗脱, 收集洗脱液离心富集卵囊和孢囊; 按照GB/T 5750.12-2006《生活饮用水卫生标准检验方法》洗脱液离心富集卵囊和孢囊。将2种试验方法离心收集到的卵囊和孢囊分别进行免疫磁分离和荧光抗体染色, 计算2种方法的回收率。结果 优化方法对隐孢子虫和贾第鞭毛虫的加标回收率分别为60%和56%, 标准方法GB/T 5750.12-2006的加标回收率分别为22%和20%。结论 优化方法对于提高生活饮用水中两虫检出率是有效的, 且可以达到降低检测成本的目的。
Objective To investigate the method for improvement of Cryptosporidium and Giardia detection rate in drinking water. Methods Standard samples were filtered with 1.2 μm microporous membrane by vacuum filtration, and then the membranes were put in phosphate buffer saline (PBS) for ultrasonic elution. The oocysts and cysts were enriched by the eluent. At the same time, the oocysts and cysts were enriched according to GB/T 5750.12-2006 “Standard examination methods for drinking water-Microbiological parameters”. The magnetic separation and fluorescent antibody staining were respectively used for the oocysts and cysts collected by the two methods, and the recovery rates of the two methods were calculated. Results The recoveries of Cryptosporidium and Giardia were 60% and 56% by optimized method, while the results were 22% and 20% according to GB/T 5750.12-2006. Conclusion The optimized method is effective to improve the detection rates of Cryptosporidium and Giardia in drinking water, and can achieve the purpose of reducing test cost.
