仙草多糖提取工艺的优化
目的 对仙草多糖碱液提取工艺条件进行优化。方法 研究碱提法提取仙草多糖的条件, 考察碱浓度、提取温度、提取时间及液料比等因素对仙草多糖得率的影响。在单因素实验的基础上, 选取碱浓度、提取时间及液料比3因素应用响应面中Box-Benhnken设计原理进行试验设计, 以仙草多糖提取率为响应值, 建立多糖提取率的二次回归方程, 得到碱液提取多糖的最佳条件。结果 仙草多糖提取的最佳浸提条件: NaHCO3 1.25%, 提取时间3 h, 液料比35:1 (V:m), 在此条件下仙草多糖提取率为8.10%。结论 碱液提取法能够提高仙草多糖的提取率。
Objective To optimize the alkali extraction conditions of polysaccharide from Mesona. Methods The alkali concentration, extraction temperature, extraction time and liquid-material ratio on the yield of polysaccharide from Mesona were investigated. On the basis of single factor experiments, 3 factors, including alkali concentration, extraction time and liquid-material ratio, were chosen to design the experiment by using the Box-Benhnken design principle in response surface. Using the extraction rate of polysaccharide as the response value, the quadratic regression equation of extraction rate of polysaccharide was established, and the optimum conditions of extracting polysaccharide from alkali solution were obtained. Results The optimum conditions of extraction of polysaccharide form Mesona were as follows: NaHCO3 was 1.25%, extraction time was 3 h, and ratio of liquid-material was 35:1 (V:m). Under the optimal conditions, the yield of polysaccharide was 8.10%. Conclusion The established alkali extraction method can increase the extraction rate of polysaccharide.
