Objective To develop a determination method for β-Carotene in fresh tea by high performance liquid chromatography (HPLC). Method The β-Carotene was extracted from fresh tea leaves by acetone. A SunFireTM C18 column (150 mm×4.6 mm, 5 μm) was used with the mobile phase of chloroform-acetonitrile solution (9:1, V:V). The flow rate was 1.0 mL/min, wavelength was 450 nm and the column temperature was 35 ℃. Result This method showed a good linearity (r=0.99997) for β-Carotene (ranging from 9.5×10-3 to 0.76 μg) and the recovery of β-Carotene was from 97.32% to 101.26%. Conclusion This method is precise, accurate and suitable for the determination of β-Carotene in fresh tea leaves.
