响应面法优化养殖大黄鱼蛋白质提取工艺
目的 优化养殖大黄鱼蛋白质提取的工艺条件。方法 通过筛选试验, 以样品提取液中总蛋白的SDS-PAGE电泳图谱的条带数和清晰度为指标, 得到养殖大黄鱼蛋白质的最适提取液。采用Plackett-Burman(PB)试验和最陡爬坡试验对提取过程中的影响因素进行评估, 然后以提取液pH、提取液浓度和液固比为自变量, 以样品提取液中的总蛋白含量为响应值, 利用响应面分析法研究各变量及其交互作用对总蛋白含量的影响。结果 选择2% SDS溶液为最佳提取液, 得到养殖大黄鱼蛋白质提取的最佳工艺条件如下: 提取液为3.50 g/100 mL的SDS溶液, pH值为8.3, 液固比为17.5:1, 提取温度为25 ℃, 提取时间为2 h。在此条件下, 样品提取液中的总蛋白含量可高达92.23%。结论 采用本研究得到的工艺条件可以实现对大黄鱼肌肉蛋白质的高效提取, 为大黄鱼蛋白质的相关研究及大黄鱼的工业化生产提供理论依据。
Objective To optimize the extraction process of cultured Pseudosciaena crocea protein. Methods Using the band number and clarity of SDS-PAGE electrophoretogram of total protein in sample extract as indexes, the optimum extract was obtained by screening test. Influencing factors in the extraction process were evaluated using Plackett-Burman (PB) design and the steepest ascent test. Then the effects of the variables and their interaction on the total protein content were investigated by response surface methodology with the pH value of extract, extract concentration and liquid-solid ratio as independent variables and total protein content in sample extract as response. Results The 2% SDS solution was selected as the best extraction solution, and the optimum extraction process conditions of cultured Pseudosciaena crocea protein were as follows: the extract was SDS solution (3.50 g/100 mL), pH value was 8.3, liquid-solid ratio was 17.5:1, extraction temperature was 25 ℃ and extraction time was 2 h. Under the above conditions, total protein content in sample extract could be as high as 92.23%. Conclusion The efficient extraction of Pseudosciaena crocea muscle protein can be realized using the process conditions obtained by this study, which can provide theoretical basis for the related researched and industrial production of Pseudosciaena crocea.
