苯甲酸钠对大鼠神经系统损伤的影响
目的 探讨食品防腐剂苯甲酸钠对大鼠神经系统的损伤机制。方法 选取40只健康SPF级SD大鼠, 分为4组, 每组10只, 以苯甲酸钠低、中、高剂量组(1457、2914和4371 mg/kg)分别喂养, 连续30天。处死后脑组织经前处理制成匀浆后, 采用定磷比色法测定Na+-K+-ATP 酶、Ca2+-Mg2+-ATP酶的活性, 探讨苯甲酸钠对大鼠脑线粒体的损害; 以苄胺为底物, 经环己烷提取, 于242 nm下测定吸光度, 继而测定出单胺氧化酶(MAO)活性; 匀浆上清液中加入巯基显色剂进行比色定量, 于412 nm下测其吸光度从而测定乙酰胆碱酯酶(AChE)的活力; 匀浆上清液中加入1.00 mL碱性羟胺, 加入0.5 mL 4 moL/L盐酸及0.50 mL 10%三氯化铁, 于530 nm 处比色, 得出乙酰胆碱(ACh)的含量。结果 苯甲酸钠影响大鼠体重的增长和一般生长发育, 可降低大鼠脑组织中ATP酶、MAO及AChE活性, 增加ACh含量。结论 苯甲酸钠在一定程度上能造成大鼠脑组织神经系统的损害。
Objective To investigate the damage mechanism of sodium benzoate on the nervous system of rats. Methods Forty healthy SD rats of SPF level were divided into 4 groups, and fed with low, medium and high dose of sodium benzoate (1457, 2914, 4371 mg/kg) for 30 days. After homogenate pre-treatment of brain tissue, the enzymatic activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase were detected by phosphorus colorimetric method, so as to investigate the damage of sodium benzoate on the mitochondria of rat brain. Using benzylamine as substrate, the activity of monoamine oxidase (MAO) was obtained by determination of absorbance at 242 nm after cyclohexane extraction, and the activity of acetylcholinesterase (AChE) was also obtained by determination of absorbance at 412 nm after adding sulfhydryl reagent for coloration. The content of acetylcholine (ACh) was obtained by determination of absorbance at 530 nm after adding 1.00 mL basic hydroxylamine, 0.5 mL 4 moL/L hydrochloric acid and 0.50 mL 10% ferric chloride into homogenate. Results Sodium benzoate could affect the growth of rat weight and general growth, reduce the activities of ATP enzyme, MAO and AChE, and increase the content of ACh. Conclusion Sodium benzoate can damage the nervous system of rat brain tissue to some extent.
