SiO2@NiO固相萃取结合超高效液相色谱-质谱法监测鸡蛋中阿苯达唑兽药及其代谢产物
目的 建立一种鸡蛋中阿苯达唑(albendazole, ABZ)兽药及其代谢产物阿苯达唑亚砜(albendazole sulfoxide, ABZ-SO)和阿苯达唑砜(albendazole sulfone, ABZ-SO2)的新监测方法。方法 将目前已知的苯并咪唑类药物标准样品直接进质谱分析, 考察其二级碎裂质谱行为, 找出其共同的特征子离子。鸡蛋样品经乙腈提取浓缩后, 经5 mL 20 mmol/L磷酸盐缓冲液(pH 7.0)溶解, 再经氧化镍涂层硅胶(SiO2@NiO)萃取柱进行固相萃取, 最后采用m/z 159特征子离子进行母离子扫描监测阿苯达唑药物及其代谢产物。结果 采用本方法监测到喂食阿苯达唑药物的母鸡产的鸡蛋中的阿苯达唑残留及其代谢产物, ABZ, ABZ-SO和ABZ-SO2的检出限分别为2.2、5和 3 ng/g。结论 该方法快速、准确, 适合食品中阿苯达唑及其代谢产物的分析监测。
Objective To establish a novel method of monitoring albendazole (ABZ) and its metabolites (albendazole sulfoxide and albendazole sulfone, ABZ-SO and ABZ-SO) in eggs. Methods The fragmentation behaviour of benzimidazoles were investigated by the direct injection of benzimidazole standards for mass spectrometry (MS) analysis, and the common product ions were determined. Egg samples were extracted with acetonitrile and then concentrated, redissolved with 5.0 mL 20 mmol/L phosphate buffer (pH 7.0). And then the albendazole and its metabolites in the extracts were processed by nickel oxide deposited on silica (SiO2@NiO) solid-phase extraction. The albendazole and its metabolites which generated characteristic product ion m/z 159 were used for precursor ion scan in MS analysis. Results Using the established method, albendazole and its metabolites were detected in egg from the hens raised with albendazol. The limits of detection of ABZ, ABZ-SO and ABZ-SO2 were 2.2, 5 and 3 ng/g, respectively. Conclusion The proposed method is fast, accurate, which is suitable for monitoring albendazole and its metabolites residues in food samples.
