γ-氨基丁酸茶薄层层析检测方法的建立
目的 探索一种经济、方便快捷、有效地测定茶叶中γ-氨基丁酸(GABA)含量的薄层层析(TLC)方法。方法 利用TLC法对茶叶中的GABA进行定性及定量分析, 着重研究展开剂、显色剂、最低限量、标准样点样浓度等因素对分离效果的影响, 并将这种方法应用于GABA茶叶加工过程中初步检测茶叶中GABA含量。结果 展开剂以正丁醇︰冰醋酸︰水比例为4︰2︰1, 显色剂以0.4%茚三酮和展开剂混合使用, 点样体积为10 μL, 标准样点样浓度为0.03 mg/mL分离效果最好, 能够将GABA和谷氨酸明显分离。结论 该方法简便、快速、准确性和稳定性高。
Objective To detect γ-aminobutyric acid (GABA) in tea through a convenient way. Methods A thin layer chromatography (TLC) method was proposed after optimizing the experimental conditions, such as developing agents, color agents, the detection limit and the concentration of standard samples. The method was applied to qualitative and quantitative analysis of GABA in the process of tea, preliminary test tea GABA content. Results This method worked well and could separate glutamic acid and GABA obviously with the developing agents ratio of n-buoh︰glacial acetic acid︰water at 4︰2︰1, 0.4% ninhydrin used as color agent, the spotting volume as 10 μL and the spotting concentration of standard sample as 0.03 mg/mL. Conclusion The proposed method is simple, rapid, accurate and stable.
