利用超高压液相色谱串联三重四级杆质谱定量 检测人乳中的α-乳白蛋白
目的 建立一种新型基于多肽标准品体系的人乳α-乳白蛋白的定量检测方法。方法 从人乳α-乳白蛋白筛选出特异性多肽, 并建立对应的检测方法; 利用化学方法人工合成特异性多肽、同位素标记特异性多肽和内标。结果 本方法所选择的多肽具有高度特异性, α-乳白蛋白的检测灵敏度为8.0 mg/100 g, 精密度均小于5.22%, 回收率均在97.21%~102.45%之间。结论 本方法建立了利用特异性多肽检测人α-乳白蛋白的方法, 具有较高的准确度、灵敏度和稳定性。通过对447份人乳样品进行检测, 初步探索了人乳中α-乳白蛋白的含量范围, 为人乳化配方奶粉的华人配方奠定理论基础。
Objective To establish a method to quantification of α-lactalbumin in human milk with peptide standard system. Methods Two signature peptides for α-lactalbumin were searched and selected from the protein digestion products. The signature peptides, isotope-labelled signature peptides and internal standard peptides were also synthesized and used for the establish the quantification method. Results The limit of quantification for α-lactalbumin were 8.0 mg/100g, respectively. Reproducibility were below 5.22 % and spiked recoveries were between 97.21 – 102.45 %. Conclusions The method validation showed a high selectivity, accuracy, reproducibility and sensitivity. The analysis results of 447 human milk samples provided the theoretical basis for the Chinese infant formula.
