喹烯酮在刺参中代谢及食用安全性研究
目的 对喹烯酮在刺参体内的代谢规律进行研究、对其代谢物3-甲基喹噁啉-2-羧酸(MQCA)对刺参食用安全性影响进行科学评价。方法 采用单因素多水平梯度设计, 以单次投喂方式进行实验; 分别以含喹烯酮20、50、80 mg/kg的饲料投喂刺参幼参, 6 h后换水并清理残饵, 实验共持续144 h, 在设定时间点采集刺参样品, 用高效液相色谱法检测样品中喹烯酮及MQCA含量。用DAS 2.0及SPSS 13.0软件对数据进行处理。结果 用药期间3个浓度组刺参幼参体内喹烯酮含量均为持续升高, 在第6 h时达到峰值, 分别为238、592、902 μg/kg, 6 h后刺参体内喹烯酮含量逐渐降低, 第48 h降至检出限以下。3个浓度组MQCA分别在给药后6、6、3 h产生, 含量呈持续上升趋势, 且与饲料中添加喹烯酮浓度正相关。结论 喹烯酮在刺参体内吸收、消除均较快; 证实MQCA为喹烯酮的代谢产物; 刺参保苗期间添加喹烯酮, 对成参食用安全性没有影响, 可安全使用。
Objective The metabolism of quinocetone in Apostichopus japonicus and the impact of a metabolite 3-methyl-quinoxaline-2-carboxylic acid (MQCA) on food safety of the adult sea cucumber were studied in this paper. Methods The multilevel experiments of single factor were integrated and the results of the experiments were analyzed. Juvenile A. japonicus in test groups were given diets containing quinocetone at levels of 20 mg/kg, 50 mg/kg and 80 mg/kg. All the water was changed and the residue diet was cleared away after 6 h. The samples were collected and detected for the concentration of quinocetone and MQCA using the high performance liquid chromatography during the 144 h. The data were processed with software DAS 2.0 and SPSS 13.0. Results The concentrations of quinocetone in the test groups showed a growing tendency and peaked at the 6th h, the Cmax were 238 μg/kg, 592 μg/kg and 902 μg/kg, respectively. The concentrations were decreased gradually after the 6th h and became undetectable at the 48th h. MQCA was detected in the three test groups after 6 h, 6 h and 3 h. The concentrations of MQCA increased gradually and had a positive correlation with the levels of quinocetone. Conclusion In this study, it is proved that quinocetone is absorbed and depleted fast in A. japonicus. MQCA is one of the metabolites of quinocetone in A. japonicus, and it will produce no effects on the food safety of adult A. japonicus if quinocetone is only given to the juvenils.
